CNS-targeted drugs differ from the non-CNS-targeted molecules in their physicochemical profiles and structural properties. The former compounds have to be more rigid and to have lower TPSA and logP for being able to cross the Blood-brain barrier (BBB). Implementing the literature data,1 Ro3, and our in-house parameters, we created a library of unique compounds that meet CNS-like criteria. The current settings will be a good starting point for drug discovery projects related to CNS-active molecules.
